SP Guard™ is a ready-to-use stabilization buffer for HRP conjugates. SP Guard™ is a ready-to-use aqueous solution, containing purified SP1 as the active component. SP1 is a newly discovered plant-derived protein, which was isolated originally from Poplar trees. SP1 displays the following characteristics:
  • SP1 is resistant to a broad range of extreme conditions including elevated temperature, proteolitic digestion organic solvents and detergents. Chaperone-like activity: SP1 protects other proteins from inactivation and aggregation, and repairs damaged proteins. SP1 activity is observed at low concentrations (about 1 micro g/ml) and under extreme conditions. SPs are a new class of proteins: SP1 belongs to a newly discovered group of plant-derived proteins (SPs). SPs don't have any sequence homology with known proteins.
  • Structural characteristics: SP1 is a small protein (12.4 kDa) with self assembly properties that spontaneously form a doughnut-shaped 12-mer homo-oligomeric complex.

Protection Unit Definition, UP:

The UP is the dilution factor of an SP1 solution at a concentration of 1 mg/ml that confers 50% protection of horseradish peroxidase (HRP) activity under standard conditions.

Standard conditions are defined as those when the activity of HRP (5 nM) drops by more than 90% after incubation for 16 h at 25 oC (40 mM HEPES buffer,pH 7.5)
